Tg2576 Transgenic Mouse Model

Alzheimer`s disease (AD) is the most common cause of memory loss and dementia. The pathological characteristic of AD is based on protein aggregates in the brain that includes amyloid plaques containing amyloid-beta (Aβ) peptides and neurofibrillary tangles (NFTs) consisting of hyperphosphorylated tau.

The Tg2576 mouse is a widely used model to study the efficacy of new compounds against AD. The Tg2576 model overexpresses human APP695 with the Swedish mutation (KM670/671NL) under the hamster prion promotor resulting in elevated Aβ levels and amyloid plaques. Moreover, Tg2576 mice on the above-mentioned background carry the Pde6brd1 retinal degeneration allele that might cause light sensitivity/blindness.

The most important characteristics of Tg2576 mice are:
  • Development of Aβ plaques along with vascular amyloid
  • Neuroinflammation
  • Loss of dendritic spines

Animals also develop cognitive deficits, but analysis of this phenotype might be disturbed by an often observed enhanced aggressive and stereotypic behavior of this strain.
