{"id":7119,"date":"2023-09-18T17:54:54","date_gmt":"2023-09-18T22:54:54","guid":{"rendered":"http:\/\/scantoxneuro.vm0857.enterprisecloud.nu\/?post_type=qps_newsletter&#038;p=7119"},"modified":"2023-09-18T17:55:37","modified_gmt":"2023-09-18T22:55:37","slug":"repeated-mptp-injections-to-model-the-parkinsons-disease-phenotype","status":"publish","type":"qps_newsletter","link":"http:\/\/scantoxneuro.vm0857.enterprisecloud.nu\/kr\/qps-newsletter\/repeated-mptp-injections-to-model-the-parkinsons-disease-phenotype\/","title":{"rendered":"Repeated MPTP injections to model the Parkinson\u2019s disease phenotype"},"content":{"rendered":"<p>Repeated MPTP injections into the mouse\u2019 intraperitoneal space result in a Parkinson\u00b4s disease brain pathology shown as degeneration of dopaminergic neurons and neuroinflammation.<\/p>\n<p>Upon entering the brain, methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP) oxidizes into the highly neurotoxic compound 1-methyl-4-phenylpyridinium (MPP+) which is selectively taken up by dopaminergic neurons. Inside neurons, MPP+ disrupts mitochondrial function and causes oxidative stress, ultimately leading to the death of these neurons.<\/p>\n<p>The effect of the repeated MPTP injections can be validated histologically by measurement of tyrosine hydroxylase (TH) levels in the caudate putamen (Fig. 1A). In parallel, Iba-1 as a marker of activated microglia and GFAP as a marker of activated astrocytes are upregulated, indicative of neuroinflammation (Fig 1B, C).<\/p>\n<p>&nbsp;<\/p>\n<p><img loading=\"lazy\" decoding=\"async\" class=\"aligncenter wp-image-7120\" src=\"http:\/\/scantoxneuro.vm0857.enterprisecloud.nu\/wp-content\/uploads\/2023\/09\/Fig.-1-rep.-MPTP-1024x338.jpg\" alt=\"Fig. 1-rep. Unpaired t-test (A, B) and Mann-Whitney U-test (C); mean + SEM; n = 8 per group; *p&lt;0.05, ***p&lt;0.001.<\/p>\n<p>MPP+ is taken up by the dopamine transporter into the dopaminergic terminals in the caudate putamen and initially damages them, while dopaminergic cell somata in the substantia nigra remain intact. The latter is confirmed by results showing unchanged levels of TH and Iba-1 in the substantia nigra of repeatedly MPTP-treated mice (Fig 2A, B) pointing towards the absence of neuronal damage while GFAP levels are increased, indicative of an ongoing inflammatory process (Fig 2C).<\/p>\n<p><img loading=\"lazy\" decoding=\"async\" class=\"aligncenter wp-image-7123\" src=\"http:\/\/scantoxneuro.vm0857.enterprisecloud.nu\/wp-content\/uploads\/2023\/09\/Fig.-2-rep.-MPTP-1024x335.jpg\" alt=\"Fig. 2-rep. Unpaired t-test; mean + SEM; n = 8 per group; *p&lt;0.05.<\/p>\n<p>Evaluation of the brain\u2019s pathology caused by repeated MPTP treatment can further be evaluated using <a href=\"http:\/\/scantoxneuro.vm0857.enterprisecloud.nu\/ex-vivo-services\/molecular-biology-biomarker-analytics\/\">biochemical methods.<\/a><\/p>\n<p>Our results show that repeated intraperitoneal MPTP injections accurately model Parkinson\u00b4s disease. The observed processes and consequential pathology in the mouse brain closely resemble a typical Parkinson\u00b4s disease pathology.
